how to make 10% EDTA in 0.1M PBS? Thank you! - (Apr/09/2008 )

I am preparing tissue for the immunohistochemistry. During the procedure of decalcification, 0.1M PB containing 10% EDTA will be used.
However I can not find rezept for it online.
Is there any friend here that could tell me how to prepare this solution?

My idea is like this:
1. Take 10g EDTA powder, add 60-80ml ddH2O, add NaOH to adjust the pH to 8.0;
2. Swirl it with magnetic stir until all the powder is soluted;
3. Add 10ml 10* PBS, add ddH2O till 100ml;
4. Filter and done!
Am I right? Thank you so much to correct me or tell me the right rezept directly.

Do you mean 0.1X or 0.1M PBS?

If you mean 0.1X, you might wish to check how much 10X PBS to add, and ask yourself whether 1X might be more suitable. PBS general isn't measured by molarity.

Also, for basic things like this I hardly ever look for recipes. It's perfectly acceptable to do the calculations yourself.

I don't know what you mean by "not measured by molarity". Yes, 10*, I mean 10X. Because at first I didn't know how to make the cross here. sorry. But please tell me how to make this.

is that pb or pbs? if pb then it is just 0.1M phosphate buffer.

then your recipe should be to use 1M phosphate buffer (10x [final]).

what is the final pH? if not 8 then you will need to adjust the pH of the edta before adding the phosphate.
