chloroform resistance - (Apr/04/2008 )
I would like to precipitate proteins from a sucrose gradient with methanol:chloroform extraction method.
The standard protocol is to centrifuge at 13kg for a few minutes to precipitate the proteins.
Since I got big volumes (4/5 mL), I'm going to use 38mL polyallomer tubes from Beckman that are reported to have "marginal resistance" to chloroform: anyone used them with chloroform? Are they suited to resist it for a few minutes?
Thank you for every advice.
WHy don't you call beckman and ask for specifics?
"marginal" means that it will resist for a short time. it should be okay if you work quickly. but, i would find something else to use (eg-glass, teflon).
Because it was friday after 6pm...
I will call them tomorrow, but often the "expert" just repeats what is written in the technical manual:
I tried to find here some first hand experience.
I think I will give a try with 1 sample and see...
