Storing primers...together? - (Mar/14/2008 )

I'm going to be running qPCR using identical and unchanging quantities of forward and reverse primer in each reaction tube. I'd like to make primer aliquots with forward and reverse primers in the same tube, as this would simplify my qPCR setup. Is there any reason why I should keep my FW and REV primers in separate tubes for storage purposes?

I thought for a moment that storing them together might be conducive to primer-dimer formation, and then recalled that any dimers should be denatured at the beginning of my PCR anyway.

But it would nice to hear someone agree with me. happy.gif

We do keep a working stock of forward and reverse primers in the same tube together without any difficulties, but the 10x solution is kept separately.

Yeah, that's fine. Keep a working solution like that, just keep the stocks separate in case you want the individual primers at some point.

most companies who sell pre-validated primers do do by sending you them in the same tube... even lyophilized this is fine
