HEPES Kinase Assay Buffer - (Mar/04/2008 )

Hi,


I need to prepare an assay buffer for a Kinase Assay -- the paper i am following uses the following buffer


50 mM HEPES (pH 7.5)
1 mM EDTA
1 mM EGTA
15 mM Mg Acetate


What would I adjust the pH of this with?

i would use koh or naoh if i have to raise the pH and acetic acid if i have to lower the pH.

the mg acetate is the clue to the acid. it is present for the mg ions. the fact that they use the acetate salt indicates that they want to avoid chloride ions.

mdfenko is absolutely right; if, beside Cl-, Na+ is to avoid titrate with native Tris; then you have to regard that stock solutions of EDTA or EGTA are also to titarte with Tris
