In vivo T cell depletion - (Mar/03/2008 )

I am interested in removing T cells via an In vivo technique! I want to deplete all T cells using a monoclonal or polyclonal antibody (or any other method except irradiation or using chemotherapy) in a rate that normal mice become completely T cell depleted (like SCID mice). Does anybody have any experiences with different Ab and also the recommended dosage or injection schedule???

See whether you can get hold of Current protocols in immunology Unit 4.1, Ada M Kruisbeek. We use YTS191 (CD4) and YTS169 (CD8) hybridomas. I've seen protocols suggesting inoculation every 5 days, but we have to inoculate every three days. You need to check the depletions by FACS to ensure maximum depletion has occurred.
