Single cuvette in double beam spectrophotometer - (Feb/22/2008 )
hi!
can anybody tell me whether we can use a single cuvette (for both reference and sample alternatively) in a double beam spectrophotometer. does it work this way...............anybody out there ever tried it!!!!!!!!!!!!

Hi tray,
I always use a single cuvette in a double beam spectrophotometer. All I do is zero the spectrophotometer first, then put my blank (i.e. buffer only) in the sample beam and measure the absorbance. Then I subtract that reading from the readings I get from my samples.
