Has anybody ever used Dharmacon ON-TARGETplus SMARTpool siRNA? - (Feb/20/2008 )

I am wodering if anybody has experience in using Dharmacon ON-TARGETplus SMARTpool siRNA. I have trouble using Ambion siRNA and I may switch to Dharmacon siRNA. Thanks for sharing.

I have used the ON-TARGETplus SMARTpool siRNA against MCM5. This is an extremely stable protein and was concerned that I wouldn't be able to get an efficient knockdown but was pleasantly surprized to see that I could obtain a 90% (or better) loss of protein expression. I was very pleased at the ease and efficiency of this product and would recommend this company for siRNA needs.

Yep am using from the same company and gettin a good knockdown . I also use the dharmafect reagent . I would recommend that smile.gif

I have used them. Great results! 90% Knockdown between 24 and 96 hours.

They provide their own transfection reagents (i think they were called 1, 2 3 & 4 smile.gif, you check out all 4 using their trial kit and use the one that works best on your particular cells. I am not sure they still sell this trial kit. But by now, they should know which one works best for which cells, just call their techrep/

acceptable.
They have good control siRNAs, fluorescent tagged siRNAs etc. tagged ones are great to determine which cells are actually transfected.

I however prefer viral siRNAs these days for their prolonged knockdown capability. Some cellular phenotypes would only emerge when gene is knocked-down for a long time--especially if you have particularly stable protein.
