T-cell proliferation assay with soluble OKT3 - (Feb/08/2008 )

Does anyone have experience using soluble OKT3 instead of the usual plate bound for human primary CD4+ T-cell assay (H3 thymadine incorp)? I plan to use an anti-mouse F(ab)2 to crosslink at 5ug/ml. Can it work? At what OKT3 concentration range? Thanks so much.

Hi, we used soluble OKT3 at 1 ug/ml and goat anti mouse to cross link at 10 ug/ml..But if you want a better results, you should also stimulate with anti-CD28 at 5 ug/ml to trigger a complete signal..
