Tris tricine SDS page blotting - (Feb/01/2008 )

Hi,
I have run a tricine (10%) SDS gel and I would like to transfer it into nitrocellulose or PVDF membrane. Do I have to do the transfer immediately or can the gel be stored and transfered later? Can someone help me with that?

it is always better to transfer immediately than to take the chance that your proteins will diffuse while in storage (although there may be some who disagree).

Thanks for the answer. Can I transfer it using a wet system and with what kind of transfer buffer? Or do I have to do it using a semy-dry blotter?

you can use whatever blotting system with which you are comfortable.

semi-dry will be faster and it is good for low mw proteins but wet blotting works at least as well, if a little slower.
