Reducing temperature-dependence of probe? - (Jan/31/2008 )

The probe that we are currently using is very temperature-dependent. In fact, we can plot intensity vs. time and see curves similar to (but a reflection of) temperature vs time. Because of this, the temperature stability requirement is considerably high if smooth intensity curves are desired. Is there a manner to design probe to reduce its temperature-dependence? Probe design (molecular biology) is not my are of expertise, so any help would be much appreciated. Currently, purchase our probe from Gene Link.

Thank you all!

What do you mean by 'probe' - pH? Expression?

The probe we use is a TaqMan probe labeled with AlexaFluor 647 reporter dye and BHQ3 quencher dye. It is what allows us to monitor the fluorescence (indirectly monitoring amplicon). Is this what you are asking?
