at higher temperatures, especially for longer times, the proteins may aggregate despite the presence of sds and 2-me (or dtt).

at 65C the proteins will react with the sds and 2-me (albeit a little slower) but are less likely to aggregate.

Thanks. I've always boiled my samples before, but I'm trying at 65ºC this week since I'm having problems with my last WB, some bands are too high!
