OD 600 , confusing - (Jan/17/2008 )
i need to measure the OD 600 of the cell growth.. however, the spectrophotometer cant detect it since the concentration of the yis culture is too high..
then i dilute it by 100X. now, im confuse how to calculate back the OD 600 of the culture before dilution..
i try times the OD 600 from the diluted culture with 100. ..
but the result i get is too high from what i observe...
i had repeat three times my dilution method.....990ul medium + 10ul sample..
so, how to calculate back my OD 600??
or anything goes wrong with my method??

why don't you calculate the concentration from the OD 600 you get and then you time that concentration.
Maybe that's what you did, I understood that you timed the OD.

Have you tried not waiting as long before measuring the OD of the culture?
