ldh-L promoter primers - Lb.plantarum (Jul/12/2004 )

I am biotechnology student from Poland. I work with
Lb.plantarum in my lab. I make master thesis.I have to
construct vector with expressed gfp gene. The problem is to
find strong promoter. I heard it has strong promoter
. So there is my question: I would like use chromosomal
DNA to amplify ldhl promoter.Where can I find sequence of ldhl
gene promoter in GenBank, I would like to design primers to
amplify this promoter from chromosomal DNA of my Lb.plantarum
NCFB1193 . HOW to do it? is it possible ? Maybe there is another strong
promoter which i could use?Please, give me some tips, I'll be
very grateful.

Are you trying to amplify ldhl promoter from plantarum genome? I searched genbank and only got two sequences and no promoter region has been defined in these sequence. you may want to check the literature to see what others have used.

if you want to get the promoter sequnce from other genomes, it may be easy to search.

btw, why don't you just buy a vector with gfp gene driven by some types of promoters such as SV40, lac, etc. Some vectors even have enhancer which guarantee high level of transcription.
