antibodies testing - (Jan/07/2008 )

HI,
I received a while ago four antibodies directed against my favorite protein. But the protein seems to be much more difficult to purifie that we first expected. None of the western shown the expected signal.
I would like to test the AB in order to be sure they are working. the compagny send with the AB the synthetic peptide used to produce the AB, but the peptide are conjugated.....So a western seems to be a bad option to test the interaction.
Is there a way to test that?????

They are coupled to an affinity resin.
First I made a western with them but the boss was not happy with that.......being couple the peptides are not migrating to the good size so it seems to be a pb for him

then maybe try to think about an ELISA. I don't know if it's a good idea...

Don't you have any tagged version of your protein? Or can you make some?
