Help with SOC medium contamination - (Dec/20/2007 )

Hi there,

I just find out that my SOC medium got contaminated!! I have never seen this before-I usually keep the SOC medium at RT, work close to fire, sterilize container whenever I use it~~How SOC medium can be contaminated? How do you usually store SOC medium??

I faced that situation before when I stored my SOC at Rt and at 4C,I found that the best way is to divide ur media to small aliquots and keep in -20c and thaw them before use

We make up 500ml and then have 50 ml aliquots. We take care as to open it very briefly and ofcourse the usual sterile techniques. Sometimes a little less concentration aliquoting SOC, can lead to contamination.

Never go into sterile media bottles but instead pour it out. May be this helps.

it's the glucose. it also helps if you keep a separate stock of FS glucose and add to the aliquot you will be using that day, right before making up your tubes.

I divide my media to small aliquots and keep in -20c and thaw them before use .

I usually aliquot it (~30-50mL) and freeze it.

SOC medium is a bacterial growth medium used to maximize the transformation efficiency of cells, so it's prone to bacterial contamination (it's the glucose - the little guys love it!).

Best to aliquot and freeze the medium.
