Do you re-use SDS-PAGE running buffer? - (Dec/14/2007 )

Hi,

How do you feel about re-using SDS-PAGE running buffer? In my old lab we used it 3 times before throwing it out but the people in my new lab insist on using it only once. What are the reasons that the buffer might "go off" in between usage??

pH may change, Cl- content increased each run, which interfere with the stacking effect, possible protein contamination due to samples with LMW run-off the gel.

Here we use it twice.

with a discontinuous buffer system you will contaminate your electrode buffers with those in the gel and sample.

I don't re-use.
ph changes, electrolyte concentration changes.
If you want a nice migration I would not re-use.

we do not re-use Tris-based but HEPES-based running buffers as HEPES is more exprensive than Tris

use it once, and throw it out.

Tris based running buffer: once only

I have try to re-use it, but the gel didn't run properly.
Besides, it is cheap and easy to make... why risk it.

We never reuse the SDS buffer.

As minnie wrote, its cheap so why bother.

always make fresh buffer from a conc. buffer solution
