HepG2 death with INTERFERin transfection - (Nov/29/2007 )

Hello all,
I am doing transfection for HepG2 with 8 microliter of INTERFERin and 1 nM siRNA in 6-well plates.

Although the company reported no cellular toxicity for INTERFERin™ , but I have extensive cell death even if I change the medium after 3 h .

I will be greatful for the help.

What was your control siRNA? Did you also see same levle of toxicity? Have you optimized the ratio between transfection agent and siRNA, the total dosages of the complexes, as well as cell density?

Could the toxicity be due to siRNA? Did you have control siRNA also?
