preparation of bacterial cell lysate - how to do? (Nov/13/2007 )

Hi all,
I am trying to do a western blot with polyclonal serum against several bacterial cell lysates
i have got few bacterial strains but can someone guide me how to lyse the cells so that there is no too much protein degradation
ofcourse it depends on the strains and hence i am giving the list of few distinct bacterial species
E.coli
Mycobacteria
salmonella
shigella
etc.

centrifuge your bacterial culture, and resuspend the pellet in laemmli buffer. Boil, and load on a gel.
a small volume of culture medium is needed (OD600 around 0.5). 1-5 uL of culture medium is enough.
