Antibodies to stain vesicles/vacuoles in my cells? - (Nov/09/2007 )

I work with dystrophic skeletal muscle and we often find vesicles/vacuoles in our muscle tissue. We have done several apoptosis tests and they always have given us negative results.

Is there a protein in the vesicle/vacuole that I might be able to stain for? Perhaps something in the membrane? We are at a loss....

There are lots of vesicular structures in a cell like endosomes (different stages) or lysosomes or even golgi body or ER. You need to do a panel of different antibodies to figure the type of vesicles.

for eg.
endosomes - rab5 or rab 7
lysosomes- LAMP 1 or 2
ER - calnexin

If you want to stain the membrane and vesicles,AM1-43,FM1-43 may work well.AM1-43 is more stable and subsequenced immunofluence staining is possible.

what kind of cells are used?

The agent is often used in the neron to study the vesicles.The agent can stain the bimolecular lamellar lipid membrane. And the direction says it is suitable for common cell lines.
