Amplifying cultures - quick question - (Oct/26/2007 )

I have 50ml of culture containg DH5 alpha transformed cells in TB +Ampicillin. I found out I actually need 100ml to do the maxiprep but this was after I had harvested the cells by spinning them down. If I resuspend the cells in the media will I be able to use these to innoculate 100ml of media? Don't want to waste my time if spinning the cells down means they can't grow anymore. Thanks for the help.

(Some persons centrifuge and wash the bacteria to get rid of beta-lactamase before innoculating in bigger volume)

Do a midi prep with 50 ml. It also yields decent amounts of DNA.
