when concentrating my protein sample my solution turns a yellow colour ? why? - (Oct/25/2007 )

hi all!
i had been having problems concentrating down my protein in an amicon conc. previously (protein precipitated) and your tips worked wonderfully!
Now however when using a buffer containing;
Tris Hcl 50mM
DTT 1mM
NaCl 150mM
Mgcl2 1mM
I GET A PALE YELLOW COLOUR when the solution is concentrated (no precipitation, thankfully!). has anyone ever noticed this before? why is this? (when protein is absent the solution remains clear)

quite often proteins are yellow.
your protein is quite concentrated. Don't worry.

may be contamination with hemoglobine, cytochromes and/or oxidized triglycerides...
