My DNA fragment in agarose gel is in 4 degree - Can I extract the fragment and use it for cloning??? (Oct/24/2007 )

Hello all, I electophoresed a restriction digestion fragment in an agarose gel and cut out the fragment and kept it in 4 degree for 1 week. Is it problem if I elute the fragment now and use this fragment for further cloning? Please give me your opinion. Thank you

Hi, I usually like to proceed to extraction immediatly after I cut an agarose band but I think there won't be any problems if you extract now. You can always check your purified DNA by running another agarose gel!

I have left the agarose gel with DNA in 4C for few days before purifying it. Shouldnt be a problem.

Yeah, I think you should be fine. I've done something similar before. As someone already said, if you are at all concerned about whether the fragment is okay, run another gel of the purified fragment and check it.

Ditto to all the above. DNA is pretty tough stuff. I've abused it in countless ways over the years but usually end up with what I want. I wish I could say the same for RNA.
