trypan blue - (Oct/22/2007 )

Hi all.

Can any one tell me how RBC look like under the stain of trypan blue?
Would it be look like white blood cell? And is there an easy way to haemolyse the RBC?? maybe by using acetic acid??

I haemolyse RBC with a short cold incubation (5 min) in 0.17M ammonium chloride, and after centrifugation at 200g 5 min to discard supernatant

Red Cells are round cells much smaller than the lymphocytes.

To lyse RBC, I do ACK lysis. There are a few threads with discussion on ACK; if U look at them it will be helpful.

In short, I pellet the cells and resuspend the cells in around 3-5ml of ACk Lysis Buffer (room temperature) and incubate for not more than 3 minutes with gentle mixing once or twice. Then, I was the cells with medium or PBS once and resuspend in medium.
