Cloning without any digestion and ligation prosedures - GATEWAY Cloning Technology (Oct/15/2007 )

Hi,
I recently heared about this technique, I would apreciate it if anybody could send me more information.

Invitrogen offers a modular assembly system of ORF´s which can directly ligated to various of their vectors; I also thought about it to use it but was too expensive for me...

You can check invitrogen's info on the matter, I think they were the ones to patent the technology:

It is quite handy as you can skip the digestion/ligation bits, although first you need to clone your insert on what they call 'entry vector'. From then on it's really straightforward, but it's only worth it if you're planing on using your insert for stuff that requires different plasmids. If you're only going to use one plasmid then this won't help you much.

The whole process is based on the recombination of specific sequences between the 'entry vector' and the 'destination vector', which is basically the vector you'll work with - for protein overexpression, plant transformation, etc, etc...

If you search any jounal database you'll find lots of applications for the technology. There are currently 'destination vectors' for loads of different uses.
