polyacrylamide gel for DNA analysis - (Oct/03/2007 )

Hi, i need to separate two DNA bands, of size 150 bp and 160 bp. I'm planning to use polyacrylamide gels after several failed attempts with agarose gels. I need the recipe of this gel and electrophoresis condition. Thank you!

Are you sure you can't cut one but not the other with a restriction enzyme? That would make it trivial.

The two bands which I want to separate is actually products of restriction digestion and their visualisation is very critical. Hence, I cannot cut one of them into even smaller fragments.

Another question. In electrophoresis, what does 1 V/cm mean? Does it mean for a gel with length (in the direction of band migration) of, for example, 15 cm, I should use 15 V?

Generally for a 5% polyacrylamide gel, what voltage should I use to separate my dsDNA of sizes 150 bp and 160 bp?
