Why the heck my PBS solution is claudy? - I just made PBS adjusted to pH 7.5 (Sep/12/2007 )

I used the regular recipe
80gNaCL
2.0gKCL
14.4g Na2HPO4
2.4g KH2PO4

adjusted to pH 7.5 and up to 1L (10x)
I stirred it but it didn't dissolved completely, so
I autoclaved it but it didn't dissolved completely still, it's cloudy!!!!

I'm puzzled
should I stirred it again until dissolved and then autoclave it again?? or should I make a new one??? wacko.gif mad.gif

You should make it again, perhaps you made a mistake at some reagents. Buying some tablet from Sigma is convenient.

Just curious...
Have you misread the label of the chemical bottle?
It is quite easy to confuse Na2HPO4 with KH2PO4.

Anyway, you may wish to remake the PBS buffer again.

Ok,,, I finnaly find out what was wrong.
I went in this morning to my lab to see if my PBS was finally dissolved, but it wasn't so, I was sure I did everything right, but ahaa!! I found what was wrong! The story is that my lab, instead of buying NaCl, they buy salt, you know, the salt that you put in your food. Well, that is what used, but today I went in and check on the label and that frikin salt is not NaCl, is salt plus sodium silicoaluminate. No wonder why I couldn't get my stuff to dissolve!!! So, I got NaCl and guess what?? problem solved!!!
In a matter of minutes I had clear 10xPBS!

So, I'm sure you should all know that , but just in case, people please! never try using regular salt instead of NaCl unless you are making LB media.
