Hey Guys,

Please can anyone tell me what the purpose of the following buffer components are? I want to set up a transport assay, measuring intracullar pH changes using a dye. Just really curious as to what the components are for.. please help!!

Tyrode buffer:
140 mM NaCl, 4 mM KCl, 0.2 mM CaCl2, 1 mM MgCl2, 10 mM HEPES, and 10 mM D-glucose, pH 7.4

well it seems it's a buffer which mimics extracellular medium. High Na + low K+... hepes for buffering and necssity of glucose for isoosmolarity.

the salts are charged with positive and negative ions which affect transportation unsure.gif
