RNA Isolation from hydatidiform mole - (Aug/22/2007 )

hithere,

it's my 1st RNA isolation experience. i took the sample from hydatidiform mole with NaCl 0,9% that storage in -80 degree celcius about 6 months. and my result was not very good. the 260/280 ratio was 1,6 in average, even lower than that.

my question is :
1. is that my sample alright?

2. what should i aware in RNA isolation?

3. what is my mistake in RNA isolation?

The 260/280 you obtained is not that bad, but there's some protein contamination. Reducing the amount of starting material and/or increasing the volume of extraction buffer usually helps.
