shaded bands in agarose gel - (Aug/20/2007 )

I am starting in molecular area, and have some doubts...
I am testing some primers in agarose gel. The bands is not defined, it appear shaded. Can i to think the primers is not working? I need a perfect band for to conclude my primers is corrects?

Could you post a photo??? is easier to see what's the problem.

what are the sizes of your primers and what is the percentage of agarose in your gel?

most times primers are checked with page or with high percentage agarose gels.

You're not going to see a well defined primer band, It appear as a dot. You sould use an acrilamide gel if you want to see them better.

But first, why you need to run your primers???

We almost never do!!!

My primers sizes is 100 up to 202 pb. The percentagem of agarese is 1.5%. I amtrying to post a photo for to show better.

Then, I don't use acrilamide gel, because my fragments has size since 100 up to 202 pb.
I run my primers for to test they. I want to know if they are correct and if will to amplification. I think this is the better way...

Then, use a 3 or 4 % agarose gel. You'll have a better resolution.

By the way, remember that primers are single stand and the leadder is doble stand, so same size run different.
