lysing cells on matrigel - (Aug/17/2007 )

I am working with HUVECs on matrigel to assay for tube formation. Has anyone ever lysed cells on matrigel to run a western? How do you get rid of the matrigel??

you may detach by EDTA w/o trypsine before lysis protocol; if you directly lyse in plates you should have contaminations with matrigel proteins; run a control of matrigel "lysis" w/o cells to check if this is really a considerable problem



I am not familiar with HUVECS but strongly interested in tubulogenesis: do they start with cysts? do you need thick layers of matrigel to observe tubuloformation? are HUVECS oncogenic or only immortalized epithelial cells?
