PPAR-gamma - (Aug/14/2007 )

Hello everyone,
If the PPAR-gamma is activated by its ligand, the amount of its mRNA will increase or decrease?
How to prove that PPAR-gamma was activated?
Thank you very much !

Hello,
I work in the field of PPARs and from my experience i can tell you that it's not necessary to have an increase of the mRNA levels after activation of PPARg with its ligand. You should measure the protein activity, which i expect that will be increase. Also protein levels don't have to increase.
Another way to check if PPARg was activated is to check for classical targets of PPARg with real time PCR. Their mRNAs levels usually they increase. I know that in this way you infer that PPARg activity and you don't really measure it, but it's a fast way to check it.

Goodluck,
Piki.
