mouse U6 promotor in human ES cells - (Aug/13/2007 )

Hi all,

has anyone ever used a mouse U6 promotor in human cells (in particular in hES cells) to achieve a gene knockdown via shRNAs?
I am just wondering if the transcription efficiency is high enough for a sufficient knockdown?

I am aware that there are leniviral vectors around harboring the H1 promotor or human U6 promotor, but it would be more convenient for me to use my ready cloned vectors.

Thanx in adavance...

human and mouse U6 pomoters are closely related. Moreover, transcription pol III si really homologous. So i would go for it.
