Templiphi amplification kit: How to determine the amount of DNA product - (Aug/10/2007 )

Hi,
I just started using the Templiphi amplification kit. In the protocol it says that 4hrs incubation can give 1.25-1.75ug DNA. How can I determine how much DNA I got?

Run concentration standards on an agarose gel along with you rca product. It will look like a smear instead of a band but you should get an estimate of the amouunt of DNA you have by the brightness of the smear.

Hello,
I have a question similar to this. I ran my Templiphi DNA on a gel and all of my products do not run very far into the gel. Meaning, the DNA is getting out of the wells but it stays near the top of the gel and all products appear as bright globs. I am assuming this means I have very high MW product and a lot of amplified DNA. Does this sound right to you? What percent gel do you recommend to run Templiphi DNA?
