ammonium sulfate precipitation - (Mar/24/2004 )

Can anybody tell me whether I can do ammonium sulfate precipitation after homogenizing my tissue with Triton x-100.

Or

Do I have to dialyse first to remove Triton x-100?

I am trying to purify an enzyme.

Triton is rarely removable by dialysis - sticks to the membrane. We recommend diluting the protein sample 1:2 so the effective Triton concentration goes down and then doing the ammonium sulfate precipitation. Do not use powder ammonium sulfate, use a saturated solution of the salt and cool everything down to 4C. Good Luck
