Need PLen Plasmid map for stable line set up - (Aug/06/2007 )

Hello All,

I have a gene of my interest in pLen plasmid. The problem is I do not have the plasmid map for this plasmid. I looked online to find published papers or company that sold this plasmid, but I could not find any.

A lot of my work is riding on this plasmid and the clock is ticking, so if anyone has the map or restriction sites on this plasmid or the antibiotic resistance please..please let me know.

Thanks

Could it be a pLenti plasmid?

No, this is (according to PMID Biochemistry. 1993 Jun 22;32(24):6302-6, Extracellular glutathione is a source of cysteine for cells that express gamma-glutamyl transpeptidase. Hanigan MH, Ricketts WA.
PMID: 8099811)
a pUC8 derivative with SV40 origin, SV40 enhancer, a human metalothionein promoter, and a multiple cloning site from pECE, a polyadenylation signal, and a poly A tract. pECE is referenced from Ellis 1986, Cell 45:721.
The source of pLEN is T. Y. White, California Biotechnology, Mountain View. (Actually, this is a description of pLEN-PT, a derivative of pLEN, so pLEN may not have all of these features).

Entrez and Google Scholar are your friend.

Thanks a lot phage 434, I did google and entrez, I got some info, but could not dig so deep.

This might just be enough for me to design some primers for sequencing.

Let me try and see how far I get. I will update you guys.
