RNA extraction from leukocytes - (Aug/06/2007 )

Hi all

I need to do RNA extraction from the human blood leukocytes. I get 1-5 mL of blood samples and will be needing to use the plasma for other studies.

I have heard a lot abt PAXgene blood RNA kit but I wonder if I could use it since I wold need to get plasma separated before I can extract RNA from the leukocytes. So far I have tried using RNeazy micro kit but have got very low yields of RNA from the samples.

Can anyone suggest me any better kit or method I could use to meet my objectives? blink.gif

Rits, I use the PAXgene kit for that purpose. You have to use the special tubes for your original blood collection instead of a serum or EDTA tube. You will get a much better yield of RNA, however the PAXgene tubes are VERY expensive.

If you decide to switch to this method, I would also suggest that you increase the centrifugation time for the original blood tubes.

I've used the RNeasy kit as well, but the yield is no where near the same.
