transfer buffer question - anyone ever use TBS as a transfer buffer? (Jul/18/2007 )

Hi All!

Has anyone ever heard of using plain old TBS as a transfer buffer when using nitrocellulose ( or PVDF, for that matter) ? I haven't heard of this , but some colleagues were arguing over this and I thought I'd throw it out to the bioforum crew for any potential input.

hello,

I always put 20% MeOH, EtOH or iPrOH in the buffer for transfer to nitrocellulose

there may exist a confusion between TBS and TGS...
but why not ? i don't have used it, as pH is different and buffering capacity change...
Moreover there is more salt in TBS.

too much NaCl; Towbin protocol uses Tris, glycine, SDS and methanol, doesn´t it?

yes,
i do use TBS as transfer buffer for mt westerns with 0.1%sds and 4% methanol for NC membrane. and i have had fantastic results
