fixing and staining the mammalian colonies with methylene blue for the first tim - could you please show me a protocol (Jul/15/2007 )

Dear friends,
I want to fix and stain my flask containing colonies of V79 (chinese hamster cell line) using methylene blue staining. could you please help me how to do. all the instruction i have is to use 0.4% methylene blue in 50 % methanol. does it mean 0.4 g methylene blue in 100 ml of 50% methanol made from (50% methanol/50% water)?! ..

also i have no idea how to fix the colonies on the surface of the flask. (they are adherent colonies). unsure.gif

You are right about 0.4% methylene blue in 50 % methanol.
