i need a protocl for prepare genomic DNA with out kit

what kind of genomic DNA, plant, animal or yeast? The extraction differs between the three groups.

And for what purpose, (long range PCR and southern blots), or short PCR work. Ie how fragmented can the extracted DNA be?

For genomic DNA I normaly use Trizol Reagent from Invitrogen. http://www.invitrogen.com/search/index.cfm...;,'150'

How do you want to clone a piece of genomic DNA into your vector?

I use DNAzol... sister product of Trizol. DNAzol is designed to only extract DNA.

However, Since you are extracting DNA from human cells, a quick Google search should pull up a number of protocols. Human cells are easy. A basic, EDTA-SDS-proteinase, phenol and RNAse protocol should do.
