Apyrase in pyrosequencing - (Jun/22/2007 )

Hi all,

"Step 4
Apyrase, a nucleotide degrading enzyme, continuously degrades unincorporated dNTPs and excess ATP. When degradation is complete, another dNTP is added. "

My question is, would the next added dNTP be degraded by apyrase before incorporation into the template?
Would it affect the result?

rate kinetics is the answer. The polymerase incoperate the dNTP into the DNA strand faster then the Apyrase can degrade all the dNTPs.

Basically the polymerase can snatch dNTPs faster then the apyrase can degrade the entire pool of dNTPs
