tissue preparation for human/mouse islet cells - (Jun/21/2007 )

I am going to stain human/mouse islets for insulin, glucagon and my interested gene. Before that, I need to prepare either frozen or fixed tissue from isolated human or mouse islets (not the pancreas). Anyone has this experience? optimal protocols?

I also need to prepare frozen sections for pancreas and human /mouse islets. Did you get some protocol.
I am a novice. Can I differentiate between different types of cell (alpha, beta, duct etc) in H& E. Is there any method to locate beta cells without using insulin AB.
