Urgent amp agar plate question! - Help with stupid question (Jun/20/2007 )

Hi Everyone,
I'm sure this is a stupid question but I'll ask it anyway. I have some amp plates that I made 5 weeks ago and I want to use them. I don't have time to make new plates so I was wondering if I can add more amp to the surface of the plates since they are expired?

My personal opinion would be to make new ones. Just because they are expired doesn't mean that all the amp is gone. Thus if you add more you will have an unknown amount there which may kill off your cells; even those with the resistance. Just my opinion, however. I would like to hear other people's comments on this as well.

That's what I am thinking also but I am in a bit of a hurry so I thought I'd give it a try.

There isnt any problem with that. You can just add some AMP to the surface and spread it. Of course please include both positive and negative controls.

If you have time, prepare new plates. If no, you can use them. just add some amp to the surface. I would Add ¼ or ½ of the amount recommended per plate because not all amp has been degraded. Just be sure plates are not contaminated and not too dry.

Thanks everyone for your quick replies! The original plates were 50ug/ml amp but the recommended is 50-100ug/ml so I thought I could just add another 50ug to the surface and still be in the range.

That should work great, make sure you spread it well/evenly and good luck!

Your amp will still be there after 5 weeks of storage in the fridge, as long as you made the plates under sterile conditions. Expiration dates do not mean that much, really.

you could add extra ampicillin to the plates but make sure you dilute it in enough LB so that you cover the entire plate else you will have pockets of different conc. of ampicillin.

just asking, where did you stored your amp plates???
usually i make a lot of amp plates and store it in 4c and use for months, faced no problem yet
and in case of immergency i ask my labmate to give me
