RNA contamination: how to get ride off it from extracted DNA? - (Jun/18/2007 )

Greetings,

I checked the purity of DNA on 0.8% agarose and I found that the it is contaminated with RNA. I have RNase that came with different protocol (promega) can I use this one to get ride off the RNA?

Is it possible to get ride off RNA from already extracted DNA? How?

check the purity of DNA using OD 260/280 too. if it contaminate with RNA then you will get the value under 1.7
well if you see RNA in you agarose gel ofcourse you could do a RNase treatment...
add sufficient amount of RNase in your sample and incubate it 37c for 30 min to 1 hour and check again in agarose
if the RNA vanish then you can proceed further with your sample

Yes, you can use RNase from different company. Once after RNase treatment, you have to clean up as well. Normal phenol chloroform method perhaps. wink.gif
