designing a primer - (Jun/16/2007 )

Dear fellow friends,
Please help me how to design primer(s) for site-directed mutagenesis.
Any suggestion on how may I go about it...

hi,
this is from the user Dax,

There are some good primer design programs out there, but most of them are expensive. Though, I have successfully developed site-directed primers using PrimerX, an online program. It can be found at: http://bioinformatics.org/primerx/

and this is the program I use;
http://frodo.wi.mit.edu/cgi-bin/primer3/primer3_www.cgi

good luck

-MuratA-

are you using a kit to achieve mutagenesis?

-Pearler-

Thank you very much. Best regards.

use a kit might be helpful.

go to some company's website (for eg. stratagene) which makes these kits. the site will have everything you need including programs to help design primers.
