collagen type I salt precepitation - (Jun/11/2007 )

anybody please can help me with knowing the selective salt precepitation of collagen type I.

i have been trying to extract the collagen type I from fibroblasts cell culture . but no yields. i want to see if i can precipitate the collagen....

Do a pepsin digestion (1mg/ml) in 0.5 M HOAc, inactivate the pepsin by adding NaOH to pH=8. If you only expect collagen type I, you can dialyze against 0.7 M NaCl, 0.5 M HOAC and the collagen type I is in the pellet. If you also have type II and III, these will also be in this pellet.

hi! this may be a bit late for you, but try Bateman, J (1984). has a protocol for 3H proline labelling.

ps collagen I will precipitate in 25% ammonium sulphate.

might need a more complete reference (eg- journal, volume, page) if there is any interest.
