How do you design primers of PCR-STR used in Forensic Medicine - Urgent!!!! (Jun/08/2007 )

Hi ladies and gentleman:

Anyone do forensic medicine? And how do you design primers for PCR-STR used in Forensic Medicine? Is there any tips?U know that STR loci have many short repeats so how can you design specific primers? Is there some specific approachs?!

Sorry, it is rather likely that short tandem repeats (STR) analysis isn't a common speciality here. Thus it is not so much about lack of interest, rather lack of real experties in this subject area.

However having read a few articles on STR, I believe that there are no special requirements when designing STR primers. The primers need only bind outside the STR region understudy. Much like any primer would do.

However due to the nature of STR, there is likely a few PCR tricks required to get the PCR reaction from truncating the product. And product lenght is all that is measure to match STRs profiles between individuals.
