Question......

Do we need formamide to prepare church buffer?

I have 2 protocols to prepare it.

1-SSC (20X).....12.5 ml
10% blocking reagent.........10 ml
phospahte buffer................2,5 ml
SDS.................................7%
50% formamide......... 25 ml
D.W.............................up to 50 ml


2- Phosphate buffer.....................0.25 M
EDTA....................................1mM
BSA.....................................1 %
SDS.....................................7%


The second one does not need formamide at all........it is still working the same?

my churhc buffer is
0.5M phosphate buffer
7%SDS
EDTA 10mM

no formamide in it
