cDNA cloning - (May/31/2007 )

Hi all, along time ago i bought a cDNA from Open Biosystems, this cDNA is on pCMV-SPORT6.1, now i need to take it out of this plasmid and put it in a pEYFPN1 plasmid for further adenovirus preparation, i am just wondering if i can do the cloning just by a simple restriction enzyme digestion, or because is a cDNA i need to put it in an special vector first... wacko.gif

Thank you!!!!!!

If you have the right sites in pEYFPN1, then proceed with simple digestion and ligation. Many times, one cannot find the right sites so one would clone it into a different plasmid or PCR amplify the insert and then clone it into the final vector.
