Protocol for amplification of gene in plasmid directly from transformed e. coli? - (May/27/2007 )

It's possible to PCR the gene out from E coli right? Or I still have to do mini prep? What about colony PCR? (I'm new)

Yes, when we do screening after transformation for example, we can just pick colonies, resuspend in water and use them as template for PCR without having to do miniprep. But I think for more important exps, it is better to do miniprep.
