Cloning short sequences - Does PCR can help clones upto 70 bps into a protein sequence (May/08/2007 )

This is the result I am going to share with others
I finally desinged the primer at the N-terminal with 75 bp over hang having restriction site at the start and a few bases extra, at the 3 prime I had 35 bases complementary to my protein sequence. So the total length was upto 104 bps.
I did PCR at 65 celsius annealing temperature with varying quantities of the primer. The results are very much successful with the primer concentration being double than the shorter primer (30 bps).
